[Solution] An electron beam radiation device 10 comprises: an electron beam radiation part 110 having an emission surface 111 for emitting an electron beam; an inert gas supply part 120 having a spray port for spraying an inert gas; a chamber section 130 for housing the emission surface and the spray port; a partitioning part 140 positioned in the chamber section and facing the emission surface; and a conveyance mechanism 150 configured such that a part of the object to be irradiated can be disposed in relation to a space 130a positioned between the emission surface and the partitioning part. The spray port is positioned on the emission surface side in the chamber section and is inclined from the emission surface side toward the partitioning part side, and in a state in which part of the object to be irradiated is disposed in the space positioned between the emission surface and the partitioning screen, the inert gas supply part is configured to spray the inert gas through the spray port to a position, on the partitioning part, facing the emission surface.

The invention provides methods of using chloroplast-selectable markers to detect nuclear genome editing. The methods involve selecting a genetic modification that has occurred in a nuclear genome of a photosynthetic cell; transforming a photosynthetic cell with a ribonucleoprotein complex that effects a genetic modification in the genome in the nucleus of a photosynthetic cell; transforming the photosynthetic cell with a construct that effects an insertion of a selectable marker into the plastome of a plastid of the cell; selecting for photosynthetic cells having the selectable marker inserted into the plastome; and thereby selecting for the genetic modification to the genome in the nucleus of the photosynthetic cell.
